Semax is a synthetic heptapeptide structurally derived from the (4–10) fragment of adrenocorticotropic hormone (ACTH), comprising the ACTH(4–7) core extended at the C-terminus by the Pro-Gly-Pro (PGP) tripeptide, giving the sequence Met-Glu-His-Phe-Pro-Gly-Pro. In the scientific literature it has been characterized as a melanocortin-derived peptide that has been the subject of neurochemical and neurotrophic-signaling research.
Published third-party studies have investigated the compound’s reported modulation of the brain-derived neurotrophic factor (BDNF) / TrkB system in rodent hippocampal tissue, as well as its examined influence on dopaminergic and serotonergic markers and on neurotrophin gene transcription in cerebral ischemia models. The PGP fragment has additionally been studied as a structural element that has been reported to slow enzymatic degradation in vitro.
